Does anyone know if my rolex should keep accurate time if I wind it a full 40 times, set it, then wear for the day and put it in its box for 24 hrs or so? Should it be accurate when I go to wear after that time or would I have to wear it daily for it to stay accurate?

It should be accurate, but this is a relative term. My Sub gains on average three seconds per day, when worn. If left in certain positions, it'll gain more or less. If I leave it alone, as you stated, it usually stays around +3/24 hours.

It should be within COSC specs, -4 to +6 seconds/day average rate.

It will be accurate. If I have mine off-wrist for more than a day, I go ahead and wind it again to full capacity. The winding rotor only really maintains the power reserve
