Interesting question. I only have 2, and I surprised myself when I came to my conclusion. I assumed it would be my GMT II Pepsi, because it was my first and the one I always wanted since I was a teenager. However after becoming obsessed with watches over the past year, I would have to say if I could only have one it would have to be my TT Sub.

It would have to be the platinum DD 118206.

If by some quirk of fate I were to be limited to one Rolex, I would choose my 14060M.
It's a very versatile watch that fits many scenarios well enough. I don't really anticipate that such a limitation would arise, but one never knows. After my BLNR comes in, it may change my mind, but until then, I'll go with the Sub.
