I'm wondering if the stick dial hour markers and both hands (hour and minute) are supposed to glow in the dark. If not, is that an indication the watch may not be authentic?

Well new yes, they are suppossed to glow. However over time the substance used to make the indices glow on an R serial number Rolex will lose some if not all of their illumination. This should not be an indicator of whether the Rolex is genuine.
